Targeting Glucosepane —
the Dominant Crosslink of Aging
The ReMatrix Bio AGE program is focused on developing an oral Constitutional Dynamic Compound (CDCpd) to recognize, unmask, and cleave glucosepane while preserving the native matrix.
The Challenge
& The Program
AGE (Advanced Glycation End-products) crosslinks accumulate in long-lived structural proteins, including collagen and elastin. These added covalent bridges can alter extracellular-matrix stiffness and remodeling.
Glucosepane is the dominant mature AGE crosslink measured in aged human skin collagen. Its reactive chemistry is masked in a stable ring. ReMatrix Bio’s proposed mechanism is to reopen that chemistry and then unlink the bridge.
ReMatrix Bio’s product objective is a single adaptive small molecule designed to recognize glucosepane, unmask its closed chemistry, and unlink the crosslink while sparing native collagen architecture.
The working lead setting is arterial stiffness/HFpEF. Ocular, renal, cartilage, and skin applications are expansion hypotheses that require their own evidence.
Evidence First
The current lead is a qualified computational candidate; it has not yet been synthesized or tested in a laboratory assay. Its selection is supported by 10,520 completed AGE-focused in silico evaluations, a 39-item computational characterization package, and 12 prepared laboratory protocols. All results for the current lead are computational; no physical data for this candidate have yet been generated.
The next planned work is synthesis of an 8–15-compound series and qualification of an assay using authentic glucosepane. Required evidence includes identified intermediates and direct confirmation of glucosepane crosslink cleavage, mass balance, and preservation of native collagen, enzymatic crosslinks, and the protein backbone.
A Relevant Track Record
In 2001, a prior version of the proprietary platform (view Approach) was used under contract with a biopharmaceutical AGE company to design a separate AGE-cleaving compound. The company synthesized it, confirmed in vitro AGE activity in its primary assay, and subsequently obtained patent protection covering one or more compounds from that program.
That historical result pertained to an accessible AGE model, not mature glucosepane. It establishes relevant technology and team experience—not evidence for the current lead. ReMatrix Bio is building an evidence-gated program to test selective glucosepane crosslink reversal.
